This isn't terribly scientific but on the basis that we're each "an experiment of one," let me offer my empirical data: I did the Boston Marathon at noon then boarded a flight for Singapore at 9 the next morning. That's about 25 hours of traveling, door to door.
I drank lots and moved a lot during the flight, including flexing in the seat and walking around, plus deep knee bends. No discernable problems, no extraordinary stiffness. But loads of sympathy and TLC from the flight attendants!
